The higher harmonics of the current caused by an alternating voltage applied to bilayer lipid membranes made of diphytanoyl phosphatidylcholine (DPhPC) in decane and tetradecane were measured. A universal relation between the amplitudes of harmonics was proposed and experimentally verified. This allowed the coefficients of expansion of the capacitance in even powers of voltage to be calculated for the DPhPC membrane in tetradecane; it also permitted comparison of the inhomogeneity in the thickness of the DPhPC membranes in decane and tetradecane.  相似文献

Summary A simplified method for isolating highly purified laterobasal membranes (LBM) from enterocytes is based on treatment of membranes with 8mm CaCl2 concentration in order to aggregate intracellular membrane contaminants. The resultant LBM showed an average 15-fold enrichment and constituted 8% of the original K-stimulated phosphatase in the initial crude homogenate. It showed typical LBM migration on counter-current distribution (CCD) and was essentially free of contamination with endoplasmic reticulum and Golgi membranes. This method is highly efficient and yields sufficient purified LBM to allow comprehensive analysis of enterocyte membrane events.  相似文献

Kinetic parameters of 3-(3, 4-dichlorophenyl)-1, 1-dimethyl urea (DCMU)-induced inhibition of electron transport in chloroplast thylakoids isolated from Phaseolus vulgaris L. cv. Oregon 1604 were determined from analysis of a convergent, parallel electrical circuit. Through this analogue, the apparent affinity of the purported binding site for DCMU (K1) and the relative amount of DCMU-insensitive electron transport (vmax1/vo) were obtained using a reiterative non-linear least squares curve-fitting procedure. Exposure of thylakoids to heat caused a gradual increase in K1 (or decrease in the affinity of the thylakoid for DCMU) with an apparent activation energy of 134 kJ mol−1. Tryptic susceptibility of a protein region regulating K1 also decreased gradually with exposure to 45°C, suggesting that the heat-induced increase in K1 might be due to a protein conformational change. On the other hand, thylakoid exposure to 45°C resulted in a rapid (<5 min) irreversible increase in vmaxI/vo, which was also the apparent result of a conformational change in a region of the protein which regulates this function. These results are suggestive of the existence of differential thermal sensitivities of proteins within the thylakoids and, perhaps, of different regions within a single membrane protein.  相似文献

SYNOPSIS Interacellular membrane potential and spontaneous changes associated with motile responses in the zooid of Vorticella convallaria Linnaeus were recorded by conventional electrophysiological technics. an all-or-none large transient depolarization (large pulse) occurs in association with a spontaneous contraction of the zooid. Abstract A cellulase-containing fraction present in the culture fluid of Trichoderma reesei grown on cellulose was obtained by fractionated centrifugation. The buoyant density of this fraction was D = 1.060 g/ml. Its ultrastructural properties, as detected by transmission electron microscopy, are given. The fraction consists of membrane vesicles attached to a carbohydrate polymer. This polymer is positive to Ruthenium red staining.
The effect of urea on the extraction and separation of acidic cellulases from this fraction is described. Linear gradient gels for both urea (up to 8.0 M urea) and polyacrylamide gels (up to 30%) were used to determine adequate separation conditions for isoelectric focusing (IEF) in a polyacrylamide gel matrix. The effect of urea on the extraction and separation conditions was tested by titration curves. In the presence of 6.0−8.0 M urea, the main cellulase-containing hydrolase complex (pIapp4.2) from this fraction is split into 3 isoenzymes and a further cellulase (pI 5.65).  相似文献

Outer membrane cytochromes catalyze the final reduction step of respiratory chains to electron acceptors that cannot diffuse through the outer membrane of Gram-negative bacteria. We developed an in vivo method to detect the orientation of outer membrane cytochromes via analysis of electron transfer reactions between these enzymes and riboflavin.  相似文献

Partition coefficients of the insecticide γ-1,2,3,4,5,6-hexachlorocyclohexane (trivially, lindane) were determined in model and native membranes. Partition in egg phosphatidylcholine bilayers decreases linearly with temperature, over a range (10–40°C) at which the lipid is in the liquid-crystalline state. Addition of 50 mol% cholesterol dramatically decreases partition (2100 falls to 100, at 10°C) and abolishes the temperature dependence. First-order phase transitions of dimyristoyl-, dipalmitoyl- and distearoylphosphatidylcholines (DMPC, DPPC and DSPC) are accompanied by a sharp increase in lindane partition. Apparently, the insecticide is easily accommodated in bilayers of short-aliphatic-chain lipids, since the partitions were 2450, 600 and 50 in DMPC, DPPC and DSPC, respectively, at temperatures 10 Cdeg below the midpoint of their transitions. The lindane partition sequence in native membranes is as follows: mitochondria, sarcoplasmic reticulum, myelin, brain microsomes and erythrocytes. This sequence correlates reasonably well with the relative content of cholesterol and is similar in liposomes of total extracted lipids, although the absolute partitions showed decreased values. Therefore, the presence of proteins in native membranes contributes to the insecticide partition, probably by favouring its interaction with lipids.  相似文献

Using the fluorescent anion 8-anilino-1-naphthalenesulphonate (ANS) for determining the membrane surface potential necessitates that the intrinsic affinity constant Ki for the ANS sites be known. Two methods are presented which do not rely on a determination of Ki at high ionic strength. They are respectively applied to neutral membranes (egg phosphatidylcholine liposomes) and highly charged natural ones (horse bean microsomes and liposomes from their phospholipids). The value of Ki appears to be insensitive to the level of occupancy of the sites, the KCl concentration and the pH in large ranges. Furthermore, the classical Gouy-Chapman model seems to describe correctly the whole set of data, provided apparent mean molecular areas larger than the published crystallographic ones are admitted.  相似文献

Liposomes containing purified H-2Kk will specifically stimulate generation of a secondary allogeneic cytolytic T lymphocyte response. Effective recognition was found to depend on the structure of the liposomes. Including detergent-insoluble plasma membrane matrix during formation resulted in liposomes having two- to fourfold more activity than those prepared using just lipid and H-2.  相似文献
